蓝光对体外培养视网膜色素上皮细胞分泌VEGF及PEDF的影响
he Influence of Blue Light Exposure on VEGF and PEDF secretion of A2E-containing retinal pigment epithelium cells
目的:探讨蓝光对体外培养含N- 亚视黄基- N- 视黄基乙醇胺(N-retinylidene-N-retinylethanolamine,A2E)的视网膜色素上皮( retinal pigment epithelium, RPE) 细胞分泌VEGF及PEDF的影响。方法:合成A2E,体外培养含A2E 的RPE 细胞, 蓝光(450±9nm)照射RPE细胞,光照强度50mW/mm2,细胞分组如下:A 组:接受蓝光照射10 min; B组:蓝光照射30 min;C组:蓝光照射60 min;D组:无蓝光照射。光照后检测细胞活力,酶联免疫吸附法检测血管内皮生长因子( vascular endothelial growth factor, VEGF) 和色素上皮衍生因子( pigment epithelium derived factor, PEDF)含量。结果:A、B、C 和D 组细胞活力分别为( 70.11±4.96)% 、( 49.87±6.83)%、( 31.29±5.78)%、( 90.28±1.9)%;VEGF/PEDF比值分别为1.14、10.88、19.42、0.88。同D组相比,B组和C组的VEGF/PEDF比值明显升高(P<0.05)结论:蓝光照射能够损伤含有A2E 的RPE 细胞, 导致VEGF和PEDF含量失衡,随着蓝光照射时间的延长,损伤作用加重。
Objective To investigate the effect of blue light exposure on vascular endothelial growth factor (VEGF) and pigment epithelium derived factor (PEDF) secretion of cultured retinal pigment epithelium (RPE) cells laden with the lipofuscin fluorophore N-retinylidene-N-retinylethanolamine (A2E). Methods The A2E -laden RPE cells were exposed to blue light (450±9 nm) light with A2E (30 μmol/l).The light intensity was 50mW/mm2.According to blue light exposure time, RPE cells were divided into: group A (10 min) ; group B (30 min) ; group C( 60 min) ; group D (without light). The viability of the cells was analysed, the level of VEGF and PEDF were determined by enzyme - linked immunosorbent assay. Results Cell viability of A, B, C and D group was( 70.11±4.96)% 、( 49.87±6.83)%、( 31.29±5.78)%、( 90.28±1.9)% respectively; VEGF/PEDF ratio was 1.14、10.88、19.42、0.88 respectively. Conclusions Blue light could induce VEDF/PEDF imbalance of A2E-containing RPE cells.
