温下肠腑方对Lewis肺癌PCNA, p16, CyclinD1蛋白表达的影响
Effect of Wenxiachangfufang on Lewis Lung Carcinoma in Mice and Influences to PCNA、p16 and cyclinD1
摘要
目的:观察温下肠腑方对Lewis肺癌移植瘤的抑制作用及对PCNA、p16、cyclinD1表达的影响。方法:C57BL/6小鼠皮下接种Lewis肺癌,随机分4组,分为模型组,化疗组,温下方组和综合治疗组,化疗组和综合治疗组DDPO.1mg腹腔注射3d,温下方组和综合治疗组中药灌胃10d。计算抑瘤率,免疫荧光组织化学方法,激光共聚焦显微镜观察PCNA、p16、cyclinD1的表达。结果:温下方组抑瘤率为34.5%,低于模型组(P<0.05);温下方组、综合治疗组和化疗组PCNA、cyclinD1表达明显低于模型组(P<0.05),其余各组无显著差异;结论:温阳除滞法为主的温下肠腑方有一定的抑制肺癌生长的作用,且与顺铂有协同作用,温下肠腑方可能通过对PCNA、cyclinD1的抑制,从而抑制肿瘤生长。Abstract
Objectives: This study is to observe the effect of Wenxiachangfu Decoction in inhibiting the growth of transplanting C57BL/6 mice of Lewis and the expression of PCNA, p16, and CyclinD1. Methods: C57BL/6 mice with transplanted Lewis tumor were randomly divided into four groups:the model group(group A),chemotherapy treated group(group B),Wenxiachangfu Decoction treated group (group C),chemotherapy plus Wenxiachangfu Decoction treated group (group D):group B and D was treated with DDP 0.1mg injecting in abdomen for 3 days,and groups C and D were treated with Wenxiachangfu Decoction for 10 days.Calculation the inhibition rates, Immunofluorescence histochemistry with the Laser Scanning Confocal Microscope were used to detect the expression of PCNA, p16, and cyclinD1. Results: The inhibition rates of tumor in group C was 34.5% ,the average tumor weight had significant difference with that in group A.the Laser Scanning Confocal Microscope study showed the expression level of PCNA and cyclinD1 in groups B,C and D was significantly lower than that in group A (P<0.05, P<0.05). Conclusions: Wenxiachangfufang Decoction has good effect in inhibiting the growth of lung cancer by regulating PCNA and cyclinD1 expressions. 关键词
