为了从蛋白水平研究成纤维细胞生长因子7(FGF7)及其受体(KGFR)在哺乳动物乳腺发育、泌乳和退化过程中表达定位的动态变化,本研究首次利用激光扫描共聚焦显微系统对昆明鼠乳腺中FGF7及其受体的表达进行了免疫荧光检测。结果表明:FGF7 主要定位于乳腺导管上皮细胞以及腺泡上皮细胞外周,同时在基质中有弱的表达。FGF7有几个显著高表达的时点,分别是青春期20d、青春期40d、妊娠初期、妊娠中期、泌乳初期、退化期初期以及退化末期,在退化期4d FGF7升高达到最高值。KGFR主要出现在导管上皮细胞以及腺泡上皮细胞上,在乳腺发育的个别时期细胞外基质中有弱表达。KGFR在青春期显著高表达的时点是青春期20d 、青春期60d 。进入妊娠期后,KGFR水平先略微下降,在妊娠10d 后开始显著上升,上升到妊娠16d 后略有下降。进入泌乳期后,在泌乳4d 时KGFR表达达到最高值,在泌乳10d 后KGFR一直下降,直至进入退化期。KGFR在退化期19d 开始显著上升。

o invest the dynamic changes in expression and localization of fibroblast growth factor 7(FGF7)and its receptor in mammals development, lactation and involution, immunofluorescence assay was used to detect FGF7 by a laser scanning confocal microscope firstly in mammary gland slices. The results revealed that: FGF7 mainly localized in the peripheral of mammary gland ductal epithelial cells and acinar cells , and a weak expression in matrix .The expression of FGF7 had a few notable high points: virgin 20d, virgin 40d, the early and middle of pregnancy, early lactation, and the initial and the late involution. The expression of FGF7 increased to the highest value in involution 4d among these periods.KGFR occurred mainly in the ductal epithelial cells and acinar cells, and a weak expression in matrix in the individual time. The higher expression of KGFR in virgin was 20d, 60d; In pregnancy, the KGFR level cutted down slightly first, and increased significantly after pregnancy 10d, then declined slightly after up to pregnancy 16d. In lactation, the expression of KGFR was highest in 4d and then fallen untill involution. The expression of KGFR increased significantly from involution 19d.
