HPLC同时测定大鼠血浆中的总黄芩素和总大黄酸的浓度及药动学研究
Simultaneous determination of total baicalein and rhein in rat plasma by HPLC and applied to pharmacokinetics study
目的: 建立HPLC同时测定酶水解后总黄芩素和总大黄酸的方法并应用于药动学研究。方法:以槲皮素为内标,经β-葡萄糖醛酸苷酶与硫酸酯酶水解血浆样品,液液萃取后用Welchrom-C18柱分离,流动相为乙腈-水(0.2 %磷酸,0.1 %三乙胺,v/v)梯度洗脱,检测波长为270 nm。结果:血浆中杂质不干扰样品的含量测定,黄芩素和大黄酸分别在0.027~28.080、0.043~10.400 μg/mL范围内线性良好,平均萃取回收率均大于75%,日内、日间精密度均小于8%,稳定性符合体内药物分析要求。结论:方法灵敏度高、精密度好,可应用于大鼠血浆水解样品的黄芩素和大黄酸的测定,并可用于进行黄芩苷和大黄酸的体内药动学研究。
Objective: To establish a HPLC for simultaneous determination of total baicalein and rhein after enzymatic hydrolysis and applied to pharmacokinetics study. Methods: Quercetin as internal standard, plasma samples enzyme hydrolysis with β-glucuronidase and sulfatase, then liquid-liquid extraction sample. The processed rat plasma samples were separated on a Welchrom-C18 column. The mobile phase was acetonitrile-water (0.2% phosphoric acid, 0.1% triethylamine, v/v) for gradient elution,detection wavelength was 270 nm. Results: Chromatograms showed endogenous peaks not interfering with the analysis. The calibration curves of baicalein and rhein were both in good linearity within the range of 0.027 ~ 28.080 and 0.043 ~ 10.400 μg/mL, respectively. The mean extraction recoveries were more than 75% while the precisions were less than 8% . The results of stability experiment meet certain requirements for the biopharmaceutical analysis. Conclusion: The method has high sensitivity, precision, and can be applied to determine rhein and baicalin in plasma after hydrolysis. This method also can be used for pharmacokinetics study of baicalin and rhein.
