B16黑色素瘤、RM-1前列腺癌荷瘤小鼠的尿液蛋白质组研究

尿液可以积累更多的变化并反映疾病的早期生理病理变化,如肿瘤。因此,尿液是寻找早期生物标志物的良好来源。本研究通过皮下分别注射B16、RM-1细胞建立黑色素瘤、前列腺癌荷瘤小鼠模型,收集肿瘤生长期间4个时间点的尿液样本,基于数据非依赖性采集(DIA)技术,通过液相色谱串联高分辨质谱 (LC-MS/MS)进行定量分析。与注射肿瘤细胞前相比,B16荷瘤模型共筛选到38种人同源差异蛋白质,其中18种蛋白质被报道与黑色素瘤相关、4种蛋白质在肿瘤可见前出现显著差异,RM-1荷瘤模型共筛选到14种人同源差异蛋白质,其中9种蛋白质被报道与前列腺癌相关、9种蛋白质在肿瘤可触摸前出现显著差异。两种荷瘤模型间也存在明显差异。通过以上实验与分析,尿液蛋白质组能够反映两种肿瘤发生发展的变化及早期生物标志物信息,并为相关肿瘤的临床早期诊断提供线索。
Urine can accumulate more changes and reflect early physiological and pathological changes of the disease, such as tumors. Therefore, urine is an ideal source for finding early biomarkers. In this study, melanoma and prostate cancer-bearing mouse models were established by subcutaneous injection of B16 and RM-1 cells, respectively. Urine samples were collected at four time points during tumor growth. Based on data-independent collection (DIA) technology, LC-MS / MS was used for quantitative analysis. Compared with that before the injection of B16 cells, 38 human homologous differential proteins were screened, of which 18 proteins were reported to be related to melanoma. Before the tumor was visible, there were four differential proteins and all were reported to be related to melanoma. Compared with that before the injection of RM-1 cells, a total of 14 human homologous differential proteins were screened, of which 9 proteins were reported to be associated with prostate cancer. Before the tumor was palpable, 9 proteins showed significant differences. There are significant differences between the two tumor-bearing models. Through the above experiments and analysis, the urine proteome can reflect the changes in the development and early biomarker information of the two tumors, and provide clues for the early clinical diagnosis of related tumors
