【目的】探究miR-34a对人脂肪间充质干细胞(human adipose-derived stem cells,hASCs)成骨向分化的作用及机制调控机制。 【方法】 用过表达miR-34a的慢病毒转染hASCs,进行碱性磷酸酶和茜素红染色检测其成骨能力。用实时荧光定量PCR和蛋白质印迹实验检测过表达miR-34a对hASCs中视网膜母细胞瘤结合蛋白2(retinoblastoma binding protein 2,RBP2)、转化生长因子β诱导因子2(transforming growth factor-β-induced factor homeobox 2,TGIF2)及runt相关转录因子2(runt-related transcription factor 2,RUNX2)表达水平的影响。 【结果】 过表达miR-34a使碱性磷酸酶和茜素红染色增强;使RBP2和TGIF2的表达水平下调,而RUNX2的表达水平上调。 【结论】 miR-34a可通过抑制RBP2和TGIF2相关信号通路促进hASCs成骨向分化。

Objective: To explore the effects of miR-34a on the osteogenic differentiation of human adipose-derived stem cells (hASCs) and regulatory mechanisms. Methods: After transfecting the hASCs with lentivirus overexpressing miR-34a, we performed the alkaline phosphatase staining and alizarin red staining to assess the level of osteogenic differentiation. The mRNA and protein expression of retinoblastoma binding protein 2 (RBP2), transforming growth factor-β-induced factor homeobox 2 (TGIF2) and runt-related transcription factor 2 (RUNX2) were examined by the quantitative real-time PCR and Western blotting after overexpressing miR-34a in the hASCs. Results: Overexpression of miR-34a significantly increased alkaline phosphatase expression and the matrix mineralization. MiR-34a downregulated the expression of RBP2 and TGIF2, while upregulated the expression of RUNX2. Conclusion: MiR-34a promotes the osteogenic differentiation of hASCs through regulating RBP2 and TGIF2 negatively.
