Label-free immunosensors based on microcantilevers of an atomic force microscope were fabricated for the rapid detection of clenbuterol by covalently immobilizing clenbuterol ovalbum onto the microcantilever. The performance and factors influencing the performance of the resulting immunosensors were investigated in detail by measuring the cantilever deflections using the optical reflection technique. High sensitivity and improved detection range can be obtained through optimization of detection conditions. The differential deflection signals revealed a specific monoclonal antibodies binding and were proportional to the logarithm of the clenbuterol antibodies concentrations in solution. Therefore, clenbuterol can be measured by determining the mixed reaction liquid of different concentrations clenbuterol and the fixed concentration clenbuterol antibodies. Under optimal conditions, the immunosensors exhibited a linear range for clenbuterol determination from 0.01 to 20 μg / L . The objective of this work was to widen the applications of microcantilever-based biosensors for small molecules.
