运用原子力显微镜(AFM)观测DNA分子,可以得到纳米级的DNA分子图像。DNA分子图像的质量和DNA在观测介质上的分散承度具有重要的关系。评价DNA在介质上的分散程度,进而对不同分散条件进行评价和优化,对于提高原子力显微镜DNA分子图像质量具有重要的意义。本研究依据大量观测结果,提出了成像质量评价的三个参数用以评价DNA分子图像质量。并依据上述参数对样品中DNA分子浓度、样品中Mg2+浓度以及环境相对湿度三个DNA分散条件进行研究。

tomic force microscopy (AFM) has been used in DNA structure for nanoscale images. DNA image quality is closely related to DNA disperse level on observation medium. It is significance for increasing DNA image quality of AFM by evaluation of DNA dispersion on medium and by optimization of different disperse conditions. In present study, we set three parameters for image quality evaluation of AFM by observation results and investigated three DNA disperse conditions: concentration of DNA sample, magnesium ion concentration in sample and environment relative humidity, according to the three parameters.
