分子连接性指数法和分子模拟法在农药免疫原
omparison of Molecular Connectivity Index and Molecular Modeling Applied for Pesticide Immunogen SAR Designing
以对硫磷为对象,研究了农药免疫原SAR设计方法。通过将分子连接性指数法和分子模拟法引入农药免疫原设计,计算了设计的免疫原与对硫磷之间的欧氏距离,根据欧氏距离的大小来判定分子之间的相似性,并通过平衡透析和ELISA实验对计算结果进行确证和比较。分子连接性指数擅长表征设计半抗原的立体结构而对电性结构表现不足,分子模拟法计算得到的电性参数对半抗原结构的表征则与此相反,而两种方法的结合能够很好的表现半抗原与目标化合物之间的分子相似度。采用分子连接性指数与电性参数结合的方法能够很好进行小分子农药免疫原的SAR设计。
Parathion was chosen as the object, the SAR designing method of pesticide immunogen was investigated. Methods of molecular connectivity index and molecular modeling were introduced into the immungoen designing, The relationship between the heterology degree of the immunogen and parathion had been investigated according to Euclidean Distance .Then the different computed results were validated and compared with the conclusions which came from the dialysis and ELISA experiments. The molecular connectivity index was good at depicting the steric structure of the designed haptens, whereas the index was not good enough to depict the electronic structure. However, the electronic parameters which came from the molecular modeling could depict the electronic structure rather than the steric structure.So when the two methods were combined, the new method could represent the degree of similarity between the desigened hapten and the analyte vere well. The molecular connectivity index combined with the electronic parameters could applied for the immunogen SAR designing of pesticide.
