探讨林下参(Panax ginseng C. A. Meyer)提取物对乙醇诱导的人永生角质细胞(HaCaT)氧化应激损伤的保护作用。首先建立乙醇诱导的HaCaT氧化损伤模型;建模成功后,细胞分为空白对照组,模型对照组,林下参提取物组。采用MTT比色法测定细胞活性,环境扫描电镜(SEM)研究细胞的形貌变化,原子力显微镜(AFM)研究细胞的杨氏模量、粗糙度、粘附力、高度的物理力学特性变化。不同剂量的林下参提取物组HaCaT细胞活性高于模型对照组。不同剂量下林下参提取物组的HaCaT细胞的细胞外基质的完整度明显高于模型对照组,并逐渐接近空白对照组。不同剂量的林下参提取物组的细胞高度低于模型对照组,杨氏模量、粘附力和细胞膜粗糙度都高于模型对照组。并且林下参提取物组的杨氏模量、高度、粘附力和膜粗糙度都接近空白对照组。林下参提取物对乙醇诱导的HaCaT细胞氧化损伤具有一定的保护作用。

he purpose of this study is to explore the protective effect of Panax ginseng extract on ethanol-induced oxidative stress damage in HaCaT.First, 6% ethanol was used to establish a cell oxidative damage model. After successful modeling, the cells were divided into 3 groups: group A was blank control, group B was model control, and group C was Panax ginseng extract. The cell viability was measured by MTT colorimetry, the morphological changes of cells were studied by SEM, and the Young\'s modulus, roughness, adhesion and height changes of the cells were studied by AFM. The HaCaT cell activity of different doses of ginseng extract group was higher than that of model control group. The integrity of the extracellular matrix of HaCaT cells in the different doses of group C was significantly higher than that of group B, and gradually approached group A. The height of cells in group C with different doses was lower than that of the model control group, and the Young\'s modulus, adhesion and cell membrane roughness were higher than group B. And the young\'s modulus, height, adhesion and membrane roughness of cells in group C were similar to group A. According to the results, Panax ginseng extract has a certain protective effect on the oxidative damage of HaCaT cells induced by ethanol.
