多不饱和脂肪酸(Polyunsaturated Fatty Acids PUFAs)对人类健康起到非常重要的作用,其中γ-亚麻酸(GLA)是合成前列腺素的前体。△6-脂肪酸脱饱和酶(D6D)是GLA等多不饱和脂肪酸合成途径的重要限速酶。月见草(Oenothera biennis)是生产天然GLA的重要油料作物,本研究克隆了其D6D基因家族2个成员(ObD6D1和ObD6D2)的全长cDNA和全长DNA序列,并进行了系统分析。ObD6D1和ObD6D2均没有内含子,全长序列分别为1922 bp和1752 bp,预测分别编码447和449个氨基酸,蛋白分子量分别为51.07 kD和51.16 kD,等电点分别为8.25和8.49,没有信号肽,定位于细胞质膜上,有一个Delta6-FADS-like保守域(cd03506),二级结构主要为α螺旋和随机卷曲,并预测产生了它们的三级结构。
Polyunsaturated fatty acids (PUFAs) play very important roles on human health, among which γ-linolenic acid (GLA) is the precursor for synthesizing prostaglandin. △6 fatty acid desaturase (D6D) is an important rate-limiting enzyme in synthesis pathway of GLA and many other PUFAs. Evening primrose (Oenothera biennis) is an important oil crop for producing natural GLA. This research cloned and analyzed the full-length cDNA and full-length DNA of two members of O. biennis D6D (ObD6D) gene family. The full-length sequences of ObD6D1 and ObD6D2 are 1922 bp and 1752 bp, respectively, both without intron. The predicted ObD6D1 and ObD6D2 proteins are 447 and 449 amino acids, with molecular weights of 51.07 kD and 51.16 kD, isoelectric points of 8.25 and 8.49, respectively. They have no signal peptide, and are located on the cytoplasm membrane. They both contain a Delta6-FADS-like conserved domain (cd03506). Their secondary structures are mainly composed of α-helix and random coil. Their tertiary structures were also predicted.
