代谢组学在肺癌诊断中的应用研究

目的:利用超高效液相色谱-质谱仪筛选肺癌诊断的潜在代谢标记物。方法:收集24例健康人和32例三种不同病理类型肺癌患者血样,对其进行前处理,除去蛋白,用超高效液相色谱-质谱(UPLC/Q-TOF)联用仪在正负离子条件下进行分离分析。markerlynx 软件归一,SIMCA-P+软件偏最小二乘法(PLS)进行分析,观察肺癌患者和正常人的代谢轮廓差异。结果:24例健康人和32例癌症病人得到很好的区分,正离子模式下发现7种差异代谢物质;而负离子模式下发现了8种差异代谢物质。结论:基于超高效液相色谱-质谱检测的代谢组学方法有助于筛选肺癌肿瘤标记物。

Objectives:The UPLC/Q-TOF metabonomics analysis methods is applied to screen tumor biomarkers in lung cancer.Methods:24 cases of healthy people and 32 cases of three different pathological types of lung cancer patients's plasma were collected. Plasma was excepted protein. With ultra-high performance liquid chromatography/ mass-spectrometry(UPLC/Q-TOF) for chromatographic separation, mass spectrometry detection and data acquisition. Data-processing was norma- lized used markerlynx software. The data was imported to SIMCA-P+ software for denoising. Finally, partial least squares (PLS) was employed for analysis.Results: 24 cases of healthy patients and 32 cases of lung cancer were distinguish well by metabolomics methods.On the pattern of ESI+,7 substances was found.on the pattern of ESI-,8 substances was found Conclusions:UPLC/Q-TOF metabonomics analysis methods may be used to identify lung cancer biomarkers.
