目的 研究敬钊素JZ-Ⅴ对学习记忆障碍的改善作用以及其LTP作用基础。方法 采用Morris水迷宫和避暗实验研究JZ-V对空间学习记忆能力和被动回避能力障碍的影响,采用电生理胞外记录方法研究JZ-V对海马突触传导通路的影响。结果 JZ-Ⅴ显著促进了大鼠海马部位突触传递通路LTP的诱导;继之,行为学实验显示:JZ-Ⅴ可显著改善不同痴呆动物模型的被动回避能力和空间学习记忆能力,定位航行实验中JZ-Ⅴ可显著缩短小鼠寻找安全岛的潜伏期和总游程;在搜索实验中,JZ-Ⅴ能显著增加两个学习记忆障碍模型小鼠在原平台象限游程比率。避暗试验结果显示:JZ-Ⅴ可明显减少小鼠5 min内的错误次数,并显著延长潜伏期。结论 本研究从药物作用的角度再次验证了LTP确为学习记忆成功的必要因子,上述JZ-V对LTP和痴呆症的认知促进作用为国内外首次发现。

im: The purposes of this study were to investigate the effect of JZ-Ⅴon learning and memory and long term potentiation. Methods: Morris water maze and Step-through experiments were applied to investigate the effect of JZ-V on spatial leaning and memory and passive avoidance lerning and memory, electrophysiological technique was used to deterine the effect of JZ-V on long term potentiation induced in granule cell of dentate gyrus. Results: JZ-V significantly enhanced the LTP induction. JZ-Ⅴsignificantly reduced escape latency and swimming distance in place navigation test, and it significantly prolonged passive avoidance latency and decreased the error numbers during 5 minutes in step-through test. Conclusions: The present study gave the strong evidence that learning and memory behavior is associated with long term potentiation, and this is the first report about JZ-V on learning and memory and long term potentiation.
