连作和头茬地黄根系钙离子浓度差异及钙信号抑制剂对连作缓解效应分析

钙信号是植物响应胁迫的重要系统,本课题组前期研究发现钙信号参与了地黄感知和响应连作毒害的过程。为了确证钙信号在连作毒害中的关键作用,本研究分别采用焦锑酸盐沉淀法、钙荧光染色法测定头茬和连作地黄根细胞内及根尖区域钙离子浓度。同时,利用钙信号阻断剂处理连作地黄,以验证其作为连作障碍消减药剂的可行性。结果表明连作胁迫显著促进地黄根尖、根内细胞内钙离子的积累,钙信号抑制剂(EGTA和异搏定)处理能够显著的缓解连作地黄的受害症状。本研究进一步从细胞水平上确证了钙信号介导了地黄连作障碍的形成,为彻底缓解连作障碍奠定了重要的工作基础。?????
alcium signals play important roles in plants in response to stress. Preliminarily studies have been indicated that calcium signals are involved in the response process that perceived and transduced harmfulness caused by consecutive monoculture practice. To identify roles of calcium signals in consecutive monoculture harmfulness, calcium ion concentration in the root cells of R.glutinosa were measured by using pyroantimonate precipitation and calcium fluorescent dyes. Simultaneously, calcium signal blockers were applied to its verification feasibility as agentia treated consecutive monoculture disease. As a result, consecutive monoculture stress significantly promoted the accumulation of intracellular calcium level in root tip and root cells of R.glutinosa. The heparin sodium had obvious effects for alleviating consecutive monoculture harmfulness compared to the untreated consecutive monoculture R.glutinosa. This study further proved roles of calcium signals in consecutive monoculture formation, and laid a soild foundation for thoroughgoing solving consecutive monoculture problems.?????
