桔梗耐盐性考察及盐胁迫对桔梗幼苗总皂苷积累的研究
Study on the Salt Tolerance and on the Platycodins Accumulation of the Platycodon grandiflorum Seedlings under Salt Stress
摘要 目的:考察桔梗种子和幼苗耐盐能力;考察盐胁迫方式对桔梗皂苷积累的影响。方法:采用不同浓度的NaCl处理桔梗种子和幼苗,测量种子萌发及幼苗生长过程中的生长和生理指标;使用不同浓度的NaCl,通过叶喷法和根灌法处理幼苗,考察其对幼苗总皂苷积累的影响。结果:随着NaCl 浓度的增加,桔梗种子萌发和幼苗生长受到抑制。种子发芽率、相对发芽率显著降低;幼苗也出现萎蔫现象,各生理指标中脯氨酸、MDA呈上升趋势,SOD、POD、CAT活性均不同程度地表现为先上升后下降的趋势。叶喷法较根灌法能够有效的促进桔梗皂苷的积累。结论:桔梗耐盐性较差,仅适合较低浓度的盐胁迫;采用叶喷法可在短期内增加桔梗总皂苷含量。
Objective: To explore salt tolerant ability of Platycodon grandiflorum seeds and seedlings, and the effects of salt stress mode. Methods: The seeds and the seedlings of Platycodon grandiflorum were treated with different concentrations of NaCl. Then, the growth traits index and physiologic index in the process of seed germination and seedling growth. The seedlings were treated with different salt concentration with foliar spray method and root irrigation method to study the effect on saponin accumulation of the seedlings. Results: With the increase of NaCl concentration, seed germination and the seedling growth were inhibited, the germination rate and relative germination rate were decreased; wilting phenomenon occur on the seedlings. The contents of proline, MDA showed increasing trends, and the activities of SOD, CAT and POD all presented a rising and then decreasing tendency. The method of foliar spray can effectively promote the accumulation of platycodins, compared with that of root irrigation. Conclusion: Platycodon grandiflorum's anti-salt performance is bad, and can be only suitable for low concentration of salt stress. The content of total saponins can be increased in the short time by leaf spraying method.
