GRK在恶性肿瘤病理中的作用

G蛋白偶联受体激酶(GRK)属于丝氨酸/苏氨酸蛋白激酶家族,其亚型广泛存在与人体组织中,通过特异性地使活化的G蛋白偶联受体(GPCR)发生磷酸化及脱敏,从而终止GPCR介导的信号转导通路发挥各种生理作用。新近研究还发现GRK的许多亚型与肿瘤的发生发展相关。本文结合GRK的功能作用和GRK在恶性肿瘤中的作用及潜在机制做简要综述。
G protein coupled receptors (GPCRs) are a superfamily of membrane receptors. G protein-coupled receptor kinases (GRKs) constitute a family of seven serine/threonine protein kinases that specifically recognize and phosphorylate agonist-activated GPCRs, thereby terminating the GPCR-mediated the signal transduction pathway. Newly researches found that in a variety of malignant tumors , GRKs play an important role in angiogenesis, proliferation, migration and invasion of tumor cells by regulating GPCRs signals and other signaling pathways. In this article, the role of GRKs in development of malignant tumor is reviewed in this paper.
