中药药物血清诱导骨髓间充质干细胞转化为肝细胞的研究
Effect of Chinese medicine drug serum on the differ entiation of bone mar row mesenchymal stem cells into hepatocytes
目的：探讨骨髓间充质干细胞(MSC s) 体外诱导其分化为肝细胞的可行性及中药调肝益气通络方的有效干预性。方法：分离、纯化并鉴定大鼠骨髓MSC,灌胃纤维化模型大鼠以调肝益气通络方后提取药物血清, 用含药血清，对照肝细胞生长因子（Hepatocyte Growth Factor ,HGF）诱导MSC向肝细胞的分化若干天后。观察各组细胞形态变化, MTT法测定细胞增殖,免疫组化检测甲胎蛋白（Alpha Fetoprotein ,AFP）与白蛋白（Albumin ,ALB）表达。 结果：1、各浓度含药血清在加入1，7，14天后，其细胞数量均显著多于常规培养组。2、含药血清诱导MSC分化7,14天后，高剂量组AFP与ALB的阳性细胞率均显著高于HGF组。结论：中药调肝益气通络方具有诱导MSC分化为肝细胞的能力。

Methods:Rat bonem arrow M SCs were isolated, purified and identified; normal and cholestaticsera of rats were prepared;Yiqi Tongluo Decoction orally fibrosis in rats after the extraction of drug serum; MSC drug-containing serum-induced liver cell differentiation 1 ，7 ，14 days; morphology was observed by immunohistochemistry of liver cells iconic protein alpha-fetoprotein (AFP), albumin (ALB), is detected.MTT assay cell growth curve,Results: 1, the concentration of serum containing 1,7,14 days, the number of cells than the conventional training group increased with the dosage, the time is proportional to.2. Serum induced differentiation of MSC positive cells after 7,14 days, AFP 、ALB of high dose group were significantly higher than in HGF group. Conclusion: YiQi Tong Luo Fang has the ability to induce MSC differentiation into liver cells.
