目的:动态观察复黄生肌愈创油膏对Wister大鼠糖尿病创面中TGF-β1、smad3、smad7含量变化的影响。方法:36只雄性Wister大鼠随机分为正常对照组,糖尿病对照组,糖尿病实验(复黄膏)组。采用免疫组化技术,观察不同时段创面中TGF-β1、smad3、smad7含量变化。结果:3天时,糖尿病实验组TGF-β1含量明显高于糖尿病对照组(P<0.05),但和正常对照组比较无统计学差异;糖尿病实验组smad3含量明显高于糖尿病对照组(P<0.05),但和正常对照组比较无统计学差异;smad7含量三组比较无统计学差异。11天时,糖尿病实验组TGF-β1含量明显低于正常对照组(P<0.05),但和糖尿病对照组比较无统计学差异;糖尿病实验组smad3含量明显低于正常对照组(P<0.05),但和糖尿病对照组比较无统计学差异;糖尿病实验组smad7含量明显高于正常对照组和糖尿病对照组(P<0.05)。结论:TGF-β1、smad3在糖尿病实验组创面愈合早期表达上调,表明复黄生肌愈创油膏通过促进创面生长因子的表达及其信号转导,从而起到促进创面愈合的作用;而在创面愈合晚期,TGF-β1、smad3在糖尿病实验组表达下调,smad7作为细胞内Smad信号的抑制剂表达上调,可能为复黄生肌愈创油膏促进了创面愈合的负反馈机制,及时中止成纤维细胞过度生长,防止瘢痕形成。

Objective:To explorer dynamic effects of“Compound Rhubarb Granulation-Promoting and Wound-Healing Ointment”(CRGWO) on the expression of TGF-β1,samd3,samd7 of granulation tissue of Wister rat diabetic wound Healing. Methods: Thirty-six male Wister rats were randomized into 3 groups:diabetic trial (CRGWO) group, normal control group, diabetic control group. The expression of TGF-β1,samd3,samd7 of granulation tissue of Wister rat diabetic wound were detected by immunohistochemical method in day 3 and day 11. Results: On the third day of wound healing, the expression of TGF-β1 and smad3 in diabetic trial group were higher than that of diabetic control group (P<0.05), but compare to the normal control group without statistical difference; the expression of smad7 in three groups have not statistical difference. On the eleventh day of wound healing, the expression of TGF-β1 and smad3 in diabetic trial group were lower than those of normal control group (P<0.05),but compare to the diabetic control group have not statistical difference; the expression of smad7 in diabetic trial group were higher than those of the others groups (P<0.05).Conclusion:The expression of TGF-β1 and smad3 in diabetic trial group were ascending in the early phase of wound healing which shows the CRGWO can promote wound healing via spurring the expression of certain cytokines and their signal transduction; in the late phase of wound healing, the expression of TGF-β1 and smad3 in diabetic trail group were descending, as well as the expression of Smad7, –the inhibitor of Smads in cell signal transduction, were ascending, which means CRGWO may open the inverse signal feedback of wound healing, thus prevent the scars formation by stopping the excessive cells proliferating.
