目的:研究缺氧环境下UCH-L1对恶性脑胶质瘤细胞侵袭性的影响并初步探讨其可能的分子机制。方法:采用细胞生物学、分子生物学和药理学等多种方法检测UCH-L1对恶性胶质瘤细胞的侵袭、存活、增殖能力的影响。结果:沉默UCH-L1后恶性胶质瘤细胞U251和T98G的侵袭能力明显下降,存活和增殖能力明显减弱,相关凋亡蛋白Caspase 3、PARP的表达明显增加。结论:UCH-L1通过抑制细胞凋亡蛋白表达,进而促进缺氧环境下恶性胶质瘤细胞的侵袭、增殖和存活。

im: To investigate the effects of UCH-L1 on invasion of glioma cells under hypoxia, and to explore the molecular mechanisms involved. Methods: Cell biology, molecular biology, and pharmacology approaches and methods were used in this study. Under hypoxia condition, UCH-L1 affect the invasion, cell proliferation, cell viability. Results: Under hypoxia condition, the ability of the migration, invasion, proliferation, viability of human glioma cells were decreased after silencing of UCH-L1 expression in human glioma cells U251 and T98G. The level of cleaved caspase 3 and cleaved PARP were significantly increased after silencing of UCH-L1 expression. Conclusion: Contributes to inhibition of apoptosis, UCH-L1 promotes the invasion, migration, survival and proliferation of glioma cells under hypoxia.???
