Exendin-4对胰岛移植治疗STZ诱导Nude糖尿病小鼠模型的治疗效果的影响
Effect of exendin-4 on islet transplantation to athymic mice with streptozotocin-induced diabetes
目的:探讨胰岛移植受体接受Exendin-4治疗,对减少供体数量提高移植效果的研究。方法:首先确定治愈25克Nude小鼠的最小胰岛数量为30个。将30只Nude糖尿病模型分为六个组,治疗组和对照组各三个组(n = 5),组别标注为治疗组(A、B、C)和对照组(A'、B'、C')。在治疗组和对照组各三个组中分别聚集地植入10个,20个,30个直径在180 μm ~ 200 μm的大鼠胰岛。治疗组在术后接受Exendin-4治疗;对照组接受生理盐水治疗。观察移植前后的体重、血糖值变化。结果:A组和A'组受体术后部分死亡,不进行分析。B组、C组和C'组在术后随机血糖分别于30.6±5.81天、10.8±2.59天和17.2±3.83天后降到11.1 mmol/L以下;而B'组在观察期内未达到糖尿病治愈的诊断标准。C'组有两只受体在观察期最后一周随机血糖值波动于5 mmol/L-18 mmol/L 之间。各组受体在术后体重呈下降趋势。B'组受体在观察期后1/3阶段体重保持稳定。B组受体体重低于B'组受体体重,且具有显著性差异(P < 0.05)。C'组受体体重在后2/3阶段呈上升趋势。C'组受体体重高于其他各组受体体重,且具有显著性差异(P < 0.05)。结论:小鼠肾被膜下胰岛移植配合Exendin-4治疗,能够显著提高移植后血糖值控制水平。
Objectives: The aim of these studies was to evaluate the effect of exendin-4 on graft function after islet transplantation. Methods: A cut-off rat islets number that just failed to cure diabetic nude mice was determined. We found that the number of 30 is the marginal number to cure diabetic mice. Then 30 recipients were divided into six groups (n=5), named group A, group B, group C, group A', group B', group C'. Group A, B and C were in treatment group that receiving 10, 20 and 30 islets respectively. Group A', B' and C' were in control group that recerving 10, 20 and 30 islets respectively. Recipients in treatment group were treated with exendin-4 intraperitoneally. And the recipients in control group were treated with vehicle. Recipients' blood glucose, body weight were measured. Results: In group A and group A', some of the mice became so weak or even died in the observation period, we stopped the observation in this two groups. The plasma glucose levels gradually decreased in group B、group C and group C', and reached to the normal range on postoperative day 30.6±5.8 and 10.8±2.59 and 17.2±3.83, respectively. The plasma level of the glucose in group B' did not decreased in to the normal range at the end of the observation period. In group C', the non-fasting blood glucose fluctuated between 5 mmol/L and 18 mmol/L in two recipients in the last 1/3 of the period. The BW deceased slowly in group B、 group C and group B', the BW was stable in the last 1/3 of the period in group B'. there was significant difference of the BW between group B and group B' (P<0.05). The BW in group C' decreased in the first 1/3 period but inrease in the last time, which had significant differences with other groups (P<0.05). Intraperitoneal glucose tolerance test (IPGTT) showed that only the mice in group C had a response that most close to the normal and mice in group C' had better glucose tolerance than that in group B. Conclusion: The glucose metabolism can be improved by the treatment of exendin-4 after the islet transplantation.
