硫酸化修饰对党参多糖体外抗Ⅰ型单纯疱疹病毒的影响
Effect of Sulfated modification of codonopsis pilosula polysaccharide against HSV-1
目的:用氨基磺酸法对党参多糖(CPPS-1)进行硫酸化修饰,检测硫酸化修饰前后党参多糖在体外抗Ⅰ型单纯疱疹病毒 (HSV-Ⅰ)的作用。方法:采用氨基磺酸法对党参多糖进行硫酸酯化修饰,红外鉴定产物结构,用氯化钡-明胶法测定硫酸根的取代度。以Vero细胞作为细胞模型,通过四甲基偶氮唑盐(MTT)法,观察比较硫酸化修饰前后的党参多糖的抗HSV-Ⅰ的作用。结果:红外光谱证实党参多糖硫酸酯化后羟基部分被硫酸基取代,在1290 cm-1 和 799cm-1 处显示了硫酸基的特征吸收峰,硫酸根的取代度D=0.754。CPPS-1和硫酸化党参多糖(SCPPS-1)在Vero细胞上的最大无毒浓度分别为100μg/ml和200μg/ml;在抗病毒实验中,CPPS-1预防及治疗给药组的细胞存活率均高于病毒对照组,但弱于阳性药物阿昔洛韦(ACV)组,而经硫酸化修饰后的SCPPS-1无论是预防给药还是治疗给药,细胞存活率均高于病毒对照组(P<0.01),且其预防作用与ACV相当(P>0.05),治疗效果优于ACV(P<0.05)。结论:CPPS-1对HSV-Ⅰ有一定的预防效果,但无治疗效果,而经硫酸化修饰后的SCPPS-1对HSV-Ⅰ具有较强的预防及治疗效果。
Objective: Sulfated modification was carried out for codonopsis pilosula polysaccharide (SCPPS-1) and evaluated the antiviral activitiy of SCPPS-1 on herpes simplex virus-1 (HSV-1). Method: Codonopsis pilosula polysaccharide was sulfated using a sulfamic acid method, and the ester sulfate structure was identified by Infrared spectrum, the ester sulfate level was turbidimetrically measured in barium chloride - gelatin. The inhibitory effect against HSV-1 were studied by MTT on Vero cell. Result: Infrared spectrum proved the ester sulfate of codonopsis pilosula polysaccharide hydroxyl portion was replaced by the sulfate ion, showed the sulfate ion of the characteristic absorption peak in 1290 cm-1 and 799 cm-1. The sulfuric acid sulfate substitution degree of D = 0.754. According to MTT assay, cell cytotoxic concentration of maximum non-toxic concentration (TC0) was calculated. For CPPS-1 and SCPPS-1, TC0 was 100μg/mL and 200μg/mL, respectively. The survival rates of vero were higher than control group, under two different administration (treatment to medicine group, preventive medicine group) of CPPS-1 by MTT assay, but lower than the ACV group. While under two different administration, the survival rates in SCCPPS-1 group were higher than control group(P<0.01), its preventive effect was equal to ACV(P>0.05), its therapeutic effect was better than that of ACV(P<0.05). Conclusion: CPPS-1 had some certain preventive effect on HSV-1 infection but no therapeutic effect, while SCCPPS-1 showed strong prevention and therapeutic effect.
