椭圆叶花锚matK基因PCR反应体系的建立和优化

以椭圆叶花锚(Halenia elliptica D. Don)为材料,利用CTAB法提取基因组DNA,通过单因素试验分析DNA模板、引物和2×TaqMasterMix 的浓度对椭圆叶花锚matK基因的PCR反应体系的影响,并对其反应条件进行优化。椭圆叶花锚matK基因优化的PCR反应体系:在25ul的反应体系中, DNA模板1.5ul,正反引物各1.75ul,2×TaqMasterMix为8ul。椭圆叶花锚matK基因片段的PCR反应体系的建立和优化,为椭圆叶花锚基因序列分析及遗传多样研究打下基础。
o establish and optimize the matK gene PCR reaction system for Halenia elliptica. Extracting genomic DNA of H. elliptica via an improved CTAB method,single factor analysis was performed to investigate the impacts of DNA template concentration,primer concentration,2 × Taq MasterMix concentration on matK gene PCR amplification and to optimize this system for H. elliptica. An optimal 25 μl volumes cpDNA matK PCR reaction system of H. elliptica would consisted of 1.5 ul DNA polymerase, 1.75μmol/L primer and 8 ul 2×TaqMasterMix. The establishment of PCR reaction system could be used to analyse the genetic diversity of H. elliptica.)
