oll样受体是一类重要的病原识别受体,能启动机体固有免疫应答并影响适应性免疫的产生。其中TLR7是Toll样受体家族中的一个特异性识别单链RNA的受体,它能第一时间对入侵机体的病毒产生响应并传递信号从而释放下游免疫因子进行自身防护。本文从四川白鹅组织中获取总RNA,利用RT-PCR和RACE技术得到四川白鹅TLR7的cDNA全场序列。通过测序发现该基因全长3900bp,包含一个完整的开放阅读框,编码1045个氨基酸。作为一个跨膜蛋白,通过软件预测到其信号肽位于第1到27位,跨膜区位于第836到860位,高度保守TIR结构域位于第884到1034位。对不同物种之间TLR7的氨基酸序列比较后,发现亲缘关系越接近物种之间基因同源性也越高。鹅TLR7基因的成功克隆为进一步研究其生物活性与免疫活性,揭示水禽固有免疫机制奠定了基础。

oll like receptors (TLRs), which are the so-called pattern recognition receptors, mediate the host innate immune response and further adaptive immune. TLR7 is one of the TLR family that especially binds single-strand RNA (almost ssRNA virus or bacteria) and rapidly transmit the signaling to the downstream immune factor inducting self-defense. In this study, we successfully isolated the total RNA from Sichuan white goose and cloned the full length of goose TLR7 cDNA by RT-PCR and Rapid-amplification of cDNA ends (RACE). Finally, we found that goose TLR7 mRNA is 3900bp with a 1045 aa ORF. As a transmembrane protein we predicted through online software, TLR7 has a signal peptide sited in position 1 to 27, transmembrane domain is sited in position 836 to 860, and the homology and conserved Toll/IL-1 Receptor (TIR) domain is sited in position 884 to 1034. When compared the amino acid alignments of TLR7 form different animals, we found that the more affinity between two species showed the more identity in the sequences. The successful work for goose TLR7 cloning is helpful for the further homology and immunology study and also provides a base line to uncover the mechanism of poultry innate immune.
