B-8大孔树脂吸附解吸中药复方多糖的研究

为了对中药复方免疫增强剂中的多糖进行了纯化,试验采用苯酚硫酸法进行含量测定,然后以AB-8大孔吸附树脂为分离材料。以乙醇为洗脱剂,系统研究上样液浓度、上样量、上样速率、洗脱剂乙醇浓度、洗脱速率、洗脱剂用量等参数对多糖吸附-解吸效果的影响。结果:确定各参数的最佳条件:上样液浓度5.93 mg/mL,上样量1倍量树脂体积,上样液速率为每小时2倍量树脂体积;洗脱剂乙醇浓度50%,洗脱速率每小时3倍量树脂体积,洗脱剂用量为3倍量树脂体积;其吸附率为74.3%,解吸率为93.6%。说明AB-8大孔树脂可用于对中药复方免疫增强剂多糖的纯化。
his study investigated compound immune enhancer of polysaccharides were extracted, and phenol hydrate-sulfuric for content determination,tested the absorption and desorption of Chinese medicine compounding polysaccharide with AB-8 macroporous resin.The polysaccharide was extracted by phenol-sulphoacid method,and with polysaccharide adsorption rate and desorption rate for index,to the AB - 8 macroporous resin adsorption and desorption conditions are optimized. Results showed that the sample liquid concentration 5.93 mg/mL, sample volume on the 1BV,sample speed 2 BV/h, elution agents ethanol concentration 50%, elution rate 3BV/h, elution agent volume for 3 BV ,the dynamic adsorption rate is 74.3%,and the dynamic desorption rate is 93.6%, which suggested that AB-8 macroporous resin can be used as purified polysaccharide material.
