基于PPI网络搜寻结直肠癌中枢基因
Searching for colorectal cancer central gene based on PPI network
【目的】本文旨在通过生物信息学方法基于PPI网络搜寻结直肠癌的中枢基因。【方法】为了搜寻结直肠癌的中枢基因,本文分析了三个来自Gene Expression Omnibus(GEO)数据库的基因芯片数据集(GSE21815,GSE21510,GSE113513)。首先将维恩分析得到的差异表达基因进行功能富集和通路分析,了解其聚类原因。然后通过STRING数据库构建差异表达基因的蛋白互作网络,并进行子网络模块分析,以更加精准地发掘结直肠癌发生过程的核心路径。最后,依据结合度筛选出结直肠癌发生过程中的中枢基因。【结果】本文发现了在整个蛋白互作网络中结合分数较高的前十个基因,将这十个基因确定为结直肠癌发生过程中起关键作用的中枢基因,并发现这些基因与细胞周期过程密切相关。所发现的中枢基因可以成为结直肠癌的治疗靶点。
【Objective】This paper aims to search for central genes of colorectal cancer based on PPI networks through bioinformatics methods.【Methods】To search for the central genes of colorectal cancer, this paper analyzed three gene chip datasets from the Gene Expression Omnibus (GEO) database (GSE21815, GSE21510, GSE113513). Firstly, the differentially expressed genes obtained by Venn analysis were subjected to functional enrichment and pathway analysis to understand the reason for clustering. Then, the protein interaction network of differentially expressed genes was constructed by STRING database, and sub-network module analysis was carried out to more accurately explore the core path of colorectal cancer. Finally, the hub genes were screened based on the degree of binding during colorectal carcinogenesis.【Results】This paper found the top ten genes with higher binding scores in the entire protein interaction network. These ten genes were identified as hub genes that play a key role in the development of colorectal cancer, and these genes are closely related to the cell cycle process. The discovered hub genes can be a therapeutic target for colorectal cancer..
