Isolation and determination of Water Soluble Polysaccharides from Phyllanthus Emblica

本文采用水提醇沉法提取余甘粗多糖,再经冻融分级、脱色素、除蛋白、透析、凝胶过滤等步骤分离多糖,测定了多糖的化学组成,高效体积排阻色谱法分析多糖纯度和分子量分布,建立了可同时对多糖中醛糖和糖酸进行精确分析的衍生化法,结合GC-MS研究多糖的单糖组成及摩尔比,红外光谱初步分析多糖结构信息。结果表明,提取分离的余甘多糖,纯度99.04 %,化学组成包括中性糖、糖醛酸和极少量蛋白质;含有两类多糖A和B,多糖A占93.64%,相对分子质量分布于1.3×105 Da,多糖B占6.36%,分子量分布于1.4×104 Da;红外光谱有糖醛酸的吸收峰,提示其是一种果胶多糖,以α-糖苷键为主;多糖由不同糖残基构成,半乳糖醛酸、半乳糖、鼠李糖和阿拉伯糖的摩尔比为3.21 : 6.59 : 1 : 0.23,此外,还含有较多的葡萄糖、木糖和少量的甘露糖。分离到的余甘多糖,含有α构型吡喃醛糖,是果胶多糖和其他半纤维素的混合物。

he crude polysaccharides from Phyllanthus Emblica were obtained with the water extraction and alcohol precipitation method, and then were separated by freeze-thawing, depigmenting, protein removing, dialysing and gel filtrating steps. The chemical composition was determined. The purity and molecular weight distribution of polysaccharides were analyzed by high performance size exclusion chromatography (HPSEC). The derivative method which could accurately analyzed aldose and sugar acid was built up and the monosaccharide composition and molar ratio of polysaccharides were measured by GC-MS. By infrared spectrum the polysaccharide structure was preliminarily analyzed. The purity of the polysaccharides by extraction and isolation was 99.04% and the chemical composition consisted of neutral sugar, uronic acid and a very small amount of protein. Two kinds of polysaccharides A and B were detected, accounting for 93.64% and 6.36% of the total amount and with predicted molecular weights of 1.3×105 Da and 1.4×104 Da, respectively. The infrared spectrum exhibited the absorption peak of uronic acid, which showed that it was a pectic polysaccharide mainly containing α-glycosidic bond. The polysaccharide was composed by different sugar residues, among which the molar ratio of galacturonic acid, galactose, rhamnose and arabinose was 3.21:6.59:1:0.23. In addition, there were still a lot of glucose and xylose and a small amount of mannose. The polysaccharides were successfully isolated from Phyllanthus Emblica, which contained pyran aldose of α configuration and was a mixture of pectin polysaccharide and hemicellulose.
