γδT细胞在沙眼衣原体呼吸道感染中的作用
he role of γδ T cells in mice with Chlamydia muridarum respiratory infection
本研究利用TCRδ-/-小鼠,探讨γδT细胞在小鼠沙眼衣原体呼吸道感染中的作用。分别给予WT(C57BL/6)和TCRδ-/-小鼠鼻腔接种1×103 IFU的沙眼衣原体小鼠肺炎菌株(Cm)制备沙眼衣原体小鼠肺炎模型。利用流式细胞仪检测感染不同时间点小鼠肺组织CD3+γδ+T细胞的百分率确定γδ T细胞在小鼠炎症部位的增殖。采用酶免疫法检测两种小鼠不同感染时期肺组织衣原体的繁殖。HE染色确定肺组织炎症水平。结果显示,Cm感染可诱导γδ T细胞在小鼠肺组织的增殖,CD3+γδ+T细胞在感染第7天达到高峰。与WT小鼠比较,Cm感染的TCRδ-/-小鼠虽然未见明显增高的肺组织衣原体繁殖,但体重下降更为显著,肺组织病理反应较重。结果提示,γδT细胞在Cm感染的早期阶段发挥重要的免疫保护作用。
Our research discussed the role of γδT cells in mice with Chlamydia trachomatis respiratory infection. WT mice and TCRδ-/- mice were intranasally inoculated with approproiate Chlamydia muridarum (Cm) to induce the Lung infection model. The percentage change of CD3+γδT cell in the lung was detected by flow cytometry technique to confirm the proliferation of γδT in different periods after the Cm infection. The burden of chlamydia in lung tissue was test by enzyme immunoassay. Lung sections were evaluated after staining with hematoxylin and eosin to score the degree of lung inflammation. The results show that γδT cells increased following infection of Cm and reached the peak on day 7. No apparent discrepancies were observed between WT and TCRδ-/- mice. however, lower weight and more severe pathologic alteration in lungs were observed in TCRδ-/- mice. From the above, γδT cells play an pivotal role in protective immunity during early stage of Cm infection.
