牛乳铁蛋白对仔猪抗菌肽Protegrin-1 mRNA表达的影响
Effect of Bovine Lactoferrin on Gene Expression of Antimicrobial Peptide Protegrin-1 in Weaning Piglets
通过饲养试验和体外实验探讨了不同浓度牛乳铁蛋白(bovine lactoferrin, bLF)对杜长大仔猪抗菌肽Protegrin-1基因表达的影响。在饲养试验中,选择90头平均体重9.70 kg左右的杜长大断奶仔猪,分成三个处理,分别为对照组(不添加bLF)、添加0.125%bLF组、添加0.25%bLF组。在体外实验中,分离培养仔猪骨髓细胞后,添加终浓度分别为10µg/ml、100µg/ml和1000µg/ml的bLF培养3、6、12和24 h,对照组用培养液替代。以18S rRNA为内标,用RT-PCR法研究日粮和骨髓细胞中添加不同浓度bLF对仔猪抗菌肽Protegrin-1基因表达的影响。结果显示:与对照组相比,仔猪日粮中添加bLF可以提高Protegrin-1基因的表达量,添加0.125%bLF和0.25%bLF分别提高了212.6%(P<0.05)和106.3%(P>0.05)。不同浓度bLF处理骨髓细胞3 h和6 h后,Protegrin-1的基因表达量增加,在12h和24h时,其表达量有所下降。结果表明,bLF可增强仔猪抗菌肽Protegrin-1基因的表达。
his experiment was conducted to study the effect of bovine lactoferrin (bLF) on antimicrobial peptide Protegrin-1 mRNA expression. In vivo,ninety weaning piglets (Duroc×Landrace×Large white) at an average weight of 9.70 kg were divided into three groups randomly, 30 piglets each group. The dietary treatments were :(1) basal diet(Control), (2) basal diet + 0.125% bLF, and (3) 0.25% bLF. In vitro, bone marrow cells were separated and cultured with bLF at the final concentration of 10, 100, and 1000µg/ml respectively for 3, 6, 12 and 24h. The control cells were incubated with culture medium. Taking 18S rRNA as inner control, the effect of bLF on Protegrin-1 mRNA expression was detected by semi-quantitative RT-PCR. The results showed that the expression levels of Protegrin-1 increased by 212.6%(P<0.05)and 106.3%(P>0.05) when the basal diet was added 0.125% bLF and 0.25% bLF respectively. After the bone marrow cells were treated with 10, 100, and1000µg/ml bLF for 3 and 6h, the expression levels increased at 3 and 6h but decreased at 12 and 24h. It suggested that antimicrobial peptide gene expression of Protegrin-1 could be regulated by bLF.
