dTe量子点荧光探针与DNA的相互作用研究
Study on interactions between CdTe quantum dots and DNA
本文采用传统水相合成法简便快速地合成了3-巯基丙酸修饰的CdTe量子点,基于两步法研究了Cd2+活化的CdTe量子点与DNA的相互作用。首先,通过在碱性条件下加入Cd2+对CdTe量子点进行活化,并详细研究了酸度、活化时间、Cd2+浓度等因素对活化效率的影响。然后基于DNA与Cd2+-CdTe量子点间的静电作用,可观察到随引入DNA浓度的增加量子点荧光逐步猝灭,且淬灭幅度与DNA浓度间呈现良好的线性关系(R=0.99198),可将本体系进一步用于复杂体系中DNA的检测。
In this paper, 3-MPA (3-mercaptopropionic acid) stabilized CdTe quantum dots (QDs) were prepared by direct aqueous synthesis method. The interactions of QDs with DNA were studied through two steps. First, by adding excessive Cd2+ under basic conditions, the prepared CdTe QDs was activated and thus cationic CdTe QDs was obtained. The effects of pH, activation time and Cd2+ concentration on activation efficiency were also studied in details. Basing on the electrostatic interactions between DNA and cationic Cd2+ enriched QDs, the fluorescence of QDs was quenched upon addition of DNA, and the quenched fluorescence intensity was in good linear relationship with the concentration of DNA (R=0.99198). This method developed here shown great potential for DNA detection in complex samples.
