维生素C通过调控成脂相关基因的表达影响3T3-L1前脂肪细胞成脂分化
Vitamin C Promotes Differentiation of 3T3-L1 Preadipocytes by Modulating Transcription of Adipogenesis-related Genes
目前关于Vit.C影响脂肪干细胞分化的结论尚有争议。在体外诱导动物脂肪干细胞成骨分化过程中,Vit.C是必需诱导剂之一。那么,Vit.C如何是影响前脂肪细胞成脂分化呢?本文利用小鼠前脂肪细胞系3T3-L1细胞,探讨Vit.C影响其分化的最佳浓度及在分化过程中成脂相关基因PPARγ2、CEBPs、SREBP-1和adiponectin 的转录及翻译水平的时序性表达变化特征。结果 随添加Vit.C浓度的升高,细胞内脂质含量也随之增加。但250μM和500 μM两组间无明显差异。结论 Vit.C促进前脂肪细胞分化的作用机制可能是通过对转录因子PPARγ2和C/EBPα以及脂代谢相关基因SREBP-1的表达调控实现的,进而促进生脂基因表达,促进脂肪细胞的分化。
he effect of vitamin C (Vit.C) on preadipocyte differentiation is not a result currently. In the process of inducing the osteogenic differentiation of animal stem cells in vitro, Vit.C is an essential inducer. Therefore, the molecular mechanism of role of Vit.C on adipocytes differentiation need to be evaluate. Here, we use 3T3-L1 preadipocyte cell lines to explore the optimal concentration of Vit.C and the changes of the time sequence characteristics of the PPAR gamma 2, CEBPs, SREBP-1 and adiponectin transcription and translation of the fat related genes in the differentiation process. The results showed that with the increasing of Vit.C concentration, the lipid content of the cells increased, the content of Vit.C 250 and 500μM was the highest, and there was no significant difference between the two groups. The mechanism may be through the regulation of transcription factor PPARγ2 and C/EBPαand lipid metabolism related gene SREBP-1 sequence regulation, promoting the expression of the fat gene, thereby promoting the differentiation of fat cells.
