神经生长因子及其受体在蛋鸡等级前卵泡上的表达
Expression of nerve growth factor and its receptors in the prehierarchical follicle of hens
为了研究神经生长因子(NGF)及其受体在蛋鸡等级前卵泡上的表达情况,本试验以83日龄性未成熟和230日龄性成熟蛋鸡各5只为试验动物,通过免疫组织化学方法和荧光定量PCR技术研究NGF及其受体TrkA和p75在蛋鸡卵巢和等级前卵泡上的表达定位及其mRNA表达水平。试验结果显示:NGF及其受体TrkA和p75在83日龄性未成熟蛋鸡卵巢上均未发现阳性表达细胞,但在230日龄等级前卵泡的膜细胞和颗粒细胞中均发现有阳性细胞表达。NGF及其受体的mRNA表达水平在230日龄蛋鸡小黄卵泡(SYF)上的表达量显著高于各自在小白卵泡(SWF)和大白卵泡(LWF)上的表达量。此外230日龄蛋鸡血清中的雌激素和孕激素水平都显著高于83日龄蛋鸡。综上所述,性成熟蛋鸡卵巢上均有NGF及其受体的表达,但是在性未成熟蛋鸡的卵巢上未发现,同时NGF及其受体的mRNA水平在SYF上的表达量显著高于SWF和LWF,由此推测NGF可能在蛋鸡等级卵泡的生长,发育和排卵过程中起一定调节作用。
In order to investigate the expression level of nerve growth factor(NGF)and its receptors in the prehierarchical follicle of hens,2 groups of 83 and 230 days old laying hens were examined by immunohistochemistry and fluorescence quantitative PCR analyses,each group contain 5 birds. The results demonstrated that NGF and its two receptors were not detected in the ovary of 83-days old laying hens, but expressed in thecal cells and granulosa cells of pre- hierarchical follicles of 230-days old laying hens.Expression of NGF and its receptors TrkA and p75 in small yellow follicle (SYF) of 230 days were significantly higher than that in small white follicle(SWF) and large white follicle(LWF).Furthermore ,serum levels of estrogen and/or progesterone were significantly higher in 230 days than that in 83 days.Taken together, these results indicated that NGF may play a role in the growth, development and ovulation of hierarchical follicles,and it may also affect the laying rate in different laying periods..
