低温处理烟草子房利于对二胞原胚的离体操作
Positive influence of low temperature treatment of the living ovary on in vitro operation of two-celled proembryos in tobacco
【目的】低温保存生物材料是一种常规手段,然而其机制并不明了。探讨了4度低温保存烟草子房的方法对二胞原胚不同参数的影响。【方法】对自新鲜子房和低温保存过的子房分离来的二胞原胚分别进行分离效率、活性和分裂潜力的检测。【结果】首先,4度低温处理子房5-23h后,二胞原胚的分离效率可显著提高23.47%。细胞学特征检测表明,低温处理后并不影响二胞原胚的细胞活性及极性,包括细胞器分布、生长方式、细胞壁组分分布。对二胞原胚的离体培养表明,低温处理后二胞原胚的离体分裂模式保持不变,但细胞分裂频率显著提高。【结论】低温处理烟草子房对二胞原胚的离体操作是有益的,不仅可提高分离效率,而且可以激发细胞分裂潜力以进一步发育。
Low temperature treatment of biological material is a routine method of preservation, but the mechanism for this is still largely unknown. Here the influence of low temperature treatment of living ovary on two-celled proembryo operation in vitro was explored in tobacco. The influence was mainly positive. First, the efficiency of isolation of two-celled proembryo was significantly improved by 23.47% for one isolation experiment after treatment of the ovary at 4 C for 5-24 h. Cellular characteristics were then investigated in detail and the results showed that the cell viability and polarity of two-celled proembryos including organelle distribution, growth style and cell wall component distribution, were all well maintained after cold treatment. Furthermore, in vitro cultivation of two-celled proembryos was carried out and showed that they could still divide normally, the division process paralleling to that of two-celled proembryos isolated from fresh ovary. Unexpectedly, the division frequency of two-celled proembryos was significantly improved by cold treatment. These results demonstrate that low temperature treatment of tobacco ovary is of benefit for two-celled proembryo operation in vitro, by not only improving isolation efficiency, but also triggering the cell's division potential for further cultivation.
