孕期镉暴露对子代发育及海马CA1区组织结构的影响

目的:观察孕期镉暴露对子代发育及海马 CA1区神经元形态结构的影响。 方法 通过在孕鼠饮水中添加10mg/L CdCl2的方法建立镉暴露动物模型,镉暴露时长从孕0天起至生产日结束。根据孕期镉暴露将子代分成2组,分别为正常对照组和镉暴露组。子代小鼠出生3周和5周时,用石墨炉原子吸收光谱法进行血清镉测定,观察镉暴露情况。在5周时,使用HE染色观察海马CA1区神经元的形态改变。结果: 子鼠出生时体重、身长和尾长都没有差别,但是子鼠出生后3周和5周时,其暴露组体重要低于正常组。石墨炉原子吸收光谱法发现,低剂量的饮用水镉暴露可以使小鼠血清镉含量明显上升(P<0.05)。HE染色:正常组海马神经元排列整齐、均匀,镉暴露后细胞数减少,神经元出现明显的病理变化。结论: 孕期低剂量镉暴露可以在子鼠体内蓄积,对其生长发育有一定的影响,对海马CA1区组织结构也有一定的影响。
Objective: The purpose of this study is to observe the effect of cadmium expose during pregnancy on offspring growth and the morphology of CA1 district of hippocampus. Method :We established the animal model of cadmium exposure by adding CdCl2 to the drinking water of pregnant mice and divided the offspring into 2 groups: control group and cadmium exposure group. And then we applied graphite furnace atomic absorption spectrometry to determine the concentration of serum cadmium and its exposure condition during 3 weeks and 5 weeks after delivery. HE staining was used to observe the morphological changes in neuronal CA1 district of hippocampus during 5 weeks after delivery. Result: the body weight of offspring from exposure group was lower than that in control group during 3 and 5 weeks after delivery. The result of graphite furnace atomic absorption spectrometry had showed that the concentration of serum cadmium was increased significantly compared with that in the control group (P<0.05). HE staining showed that the neuronal of hippocampus from control group were arranged neatly and well-distributed, while the amount of neuronal cells were decreased and neuronal cell distributed disorderly and pathologically changed obviously in cadmium exposure group. Conclusion: the low-dose cadmium exposure during pregnancy would result in the accumulation of cadmium in offspring of mice, and show a certain influence on development of growth and morphology of CA1 district of hippocampus.
