Effect of CDA-2 on nestin expression of human glioma cell line SWO-38
目的 研究人尿萃取物CDA-2（cell differentiation agent-2,又名尿多酸肽）对SWO-38人胶质瘤细胞nestin（巢蛋白）表达的影响及其诱导分化作用。 方法 免疫组化法检测56例胶质瘤石蜡标本nestin表达和胶质瘤恶性程度之间的相关性；光镜观察和鉴定细胞分化；RT-PCR、细胞免疫荧光、Western blotting分析CDA-2对SWO-38细胞nestin表达的影响。结果 nestin表达和胶质瘤恶性度之间呈正相关；光镜观察发现经CDA-2诱导后SWO-38细胞胞体变大、核浆比减小、突起增多，表现出向成熟的星形细胞分化的现象；CDA-2可下调SWO-38细胞nestin的表达。结论 Nestin表达和胶质瘤恶性度之间呈正相关；CDA-2可下调SWO-38细胞nestin的表达，诱导细胞分化；Nestin有可能成为胶质瘤诱导分化治疗领域新的标志物。
im To investigate the effect of CDA-2(Uroacitide), a healthy human urine extraction ,on nestin expression and cellular differentiation of human glioma cells SWO-38. Methods The correlation between nestin expression and the malignant grade has been studied in 56 gliomas by immunohistochemistry. Cellular differentiation of SWO-38 cells induced by CDA-2 was determined with light microscopy observation. The effect of CDA-2 on nestin expression of SWO-38cells was detected by immunocytochemistry, RT-PCR and Western blotting. Results There was positive correlation between nestin expression and the malignant grade in 56 gliomas . Light microscopy observation revealed that CDA-2 could induce SWO-38 cells differentiate into astrocyte with increased cytoplasm and cytoplasmic processes and decreased nucleus/cytoplasm rate. CDA-2 downregulated nestin expression in SWO-38 cells. Conclusion There was positive correlation between nestin expression and the malignant grade in 56 gliomas . CDA-2 downregulated nestin expression and induced differentiation in SWO-38 cell lines. Nestin may be a new differentiation marker of glioma.
