内源转基因是一项较新的转基因策略,即利用物种本身的基因和相关序列进行作物转基因遗传改良,可以降低异源序列带来的风险。为构建马铃薯内源转基因载体,本课题组在前期研究基础上,采用PCR扩增、酶切、连接和热激转化等分子克隆方法,利用实验室克隆获得的马铃薯类"T-DNA"元件替换双元质粒载体pBI121中的T-DNA右边界序列的策略,成功构建马铃薯内源转基因质粒载体,为马铃薯内源表达体系的建立打下重要基础。

Recently scientists propose a new strategy for?transgene named intragenesis. It is the genetic modification of a recipient plant with a natural gene from a crossable-sexually compatible-plant. This could avoid the risk of transgenes. To construc a potato intragenic plasmid, PCR amplification, restriction enzyme digestion, ligation and heat shock transformation were used to clone the potato intragenic plasmid by using a potato derived T-DNA like element 1RB to replace right border sequence of T-DNA in binary plasmid vector pBI121. It will lay an important foundation for establishment of potato endogenous expression system.
