Identifion and molecular mapping of a novl semi-dwarfing gene introgressed from Thinopyrum pontion into wheat

warfing line 31505-1, with reduced height than its common wheat parent, was derivative of common wheat and Thinopyrum ponticum. Genetic analysis revealed that dwarfism of 31505-1 was controlled by a single partial dominant gene, and was named as Rht-Thp. Based on the 425 F2 individuals derived from a cross of 31505-1/Chinese Spring and SSR, STS analyses, the reduced height gene of 31505-1 was mapped on chromosome 2DL, and a genetic map comprising about 72.8 cM was constructed with the loci order Xwmc41-Rht-Thp-Xmag4059-Xmag3596-Xcfd168-Xcfd267-Xmag1280-Xwmc18. Of the seven mapped markers, Xwmc41, Xmag4059, Xmag3596 and Xcfd168 amplified unique bands of Th. ponticum in 31505-1, which revealed presence of introgressed Th. ponticum segment in 31505-1 and thus proved reduced height gene was transferred from Th. ponticum. Considering its origin and the chromosome location, the reduced height gene of 31505-1 was novel, it would be a new source for broadening the genetic base of dwarfism in wheat improvement.
