荞麦水溶性多糖的分离纯化及其相对分子量的测定
Isolation and Purification of a water-soluble Polysaccharide from Fagopyum esculentum and Determination of its molecular weight
本文通过研究了通过热水浸提法从荞麦中提取了一种水溶性多糖,并用离子交换柱和凝胶过滤柱对荞麦多糖进行分离纯化、测定了其相对分子量。实验结果表明,荞麦多糖粗品用Savag 法脱蛋白后,采用DEAE-Sepharose Fast Flow 和Sephacryl S-400 HR 柱层析纯化,得到荞麦多糖FEP,经过凝胶柱色谱分析表明,FEP 为均一多糖。用苯酚硫酸法测定多糖的含量,用AKTA快速液相蛋白层析仪的紫外检测器在280nm监测蛋白质含量。最后根据用不同分子量的标准葡聚糖的做出的标准曲线,测定出FEP 分子量(Mw)为1720442Da。
In this paper, a water-soluble crude polysaccharide, obtained from Fagopyum esculentum by boiling-water extraction and ethanol precipitation, was fractionated by DEAE–Sepharose Fast Flow column chromatography, and purified by Sephacryl S-400 HR column chromatography. It was showed only one symmetrical peak from gel-filtration chromatography, indicating that FEP was a homogeneous polysaccharide. The total sugar content of each tube was determined by the phenol-sulfuric acid assay and the protein content was detected by the UV detector at the wavelength of 280 nm. The weight-average molecular weight (Mw) of the purified polysaccharide (FEP) was calculated to be 1720442 Da, according to the calibration curve with standard dextrans.
