UPLC-MS/MS测定姜黄素在MCF-7/Adr细胞中的亚细胞分布研究
Validated UPLC-MS/MS method for simultaneous determination of curcumin in subcellular compartments of MCF-7/Adr cells
目的 研究姜黄素在MCF-7/Adr细胞中的亚细胞分布情况。方法 建立姜黄素UPLC-MS/MS测定方法,考察姜黄素溶液与MCF-7/Adr细胞共培养不同后,在MCF-7/Adr细胞中不同亚细胞蛋白(细胞质、细胞膜、细胞核和细胞骨架)中的分布情况。结果 姜黄素浓度在5-2000 ng/mL浓度范围内线性良好,y = 0.00115x + 0.00517 (r = 0.9910);低、中、高3个浓度的回收率为99.21%、96.08%和100.45%,日间和日内精密度均低于15%。姜黄素0.5-2 h内,主要分布于细胞核、细胞膜/细胞器中,在细胞骨架和细胞浆中少有分布。结论 定量评价了姜黄素在肿瘤细胞MCF-7/Adr细胞(细胞质、细胞膜、细胞核和细胞骨架)中的定位及作用机制,为姜黄素抗肿瘤机制的研究提供了一定的实验依据。
o investigate distribution of Cur in subcellular compartments of resistant MCF-7/Adr cells, a specific and sensitive high-performance liquid chromatography-tandem mass spectrometric (LC-MS/MS) method has been developed and validated for the determination of curcumin (Cur) in intracellular compartments. Sequential extraction of four subcellular protein fractions (cytosolic, membrane/organelle, nucleic and cytoskeleton) was performed directly from MCF-7/Adr cells after incubation with 8 μg/mL Cur for 0.5, 1, 2 and 4 h, respectively. The calibration curves were linear over a concentration range of 5 - 2000 ng/mL for Cur (y = 0.00115x + 0.00517, r = 0.9910). The recoveries were 99.21%, 96.08% and 100.45% for three concentrations, respectively. The intra-day and inter-day precision was less than 15%. Cur was mainly distrubuted in nucleus and membrane (organelle) during 0.5 h and 2 h, seldom in cytosolic and cytoskeleton. In conclusion We quantitatively determin the location and mechanism of Cur in the subcellular compartment of resistant MCF-7/Adr cells. It will provide some experimental datas for the mechanism study of Cur in antitumor.
