保安族口腔健康儿童唾液微生物群落结构分析

目的:通过对甘肃保安族地区口腔健康儿童唾液微生物群落结构分析,探寻口腔健康儿童唾液中的优势微生物种群。方法:选取8例保安族口腔健康儿童,采集唾液样本,提取细菌总DNA。扩增16S rRNA基因后构建克隆文库,随机挑选400个阳性克隆子进行序列测定,对测序结果进行BLAST 对比。去除载体及嵌合体后用Mothur、Clustalx2.0等软件进行细菌群落结构多样性分析,用MEGA4.0构建系统发育树。结果:经测序筛选后得到387条合格序列;共检测到84个OTU,归属于5个门,12个纲,15个目,18个科,19个属。其中以厚壁菌门(Firmicutes, 88.60%)比例最多;19个菌属中共有6个优势菌属(检出率>1%),分别为链球菌属(Streptococcus, 70.28%)、颗粒链菌属(GranuIicatella, 6.20% )、孪生菌属(Gemella, 3.10%)、韦永氏球菌属(Veillonella, 1.81%)、奈瑟菌属(Neisseria, 1.03%)和嗜血杆菌属(Haemophilus, 1.03%),其中,链球菌属在8个样本中均出现。结论:保安族口腔健康儿童唾液微生物群落结构较为丰富;链球菌属是保安族口腔健康儿童唾液中的优势菌属,其在维持儿童口腔健康状态中的具体作用机制有待进一步研究。
Objective: To explore the dominant salivary microbiota in healthy children by analyzing microbial community structure of healthy children in Bonan of Gansu province. Methods: A total of 8 Bonan's children with healthy oral cavity were included in this study, Genomic DNA was extracted from saliva samples. 16S rRNA clone library was constructed after the 16s rRNA was amplified, then selected 400 positive clones randomly to sequences and compared the sequencing results by BLAST. After removing the vectors and chimera, the analysis of diversity of bacterial community structure was conducted by software such as Mothur, Clustalx2.0, etc. Phylogenetic tree was constructed used MEGA4.0. Results: After sequencing and screening, 387 qualified sequences were obtained. They were classified into 84 OTUs which belonging to5 phyla, 12 classes, 15 orders, 18 families, 19 genera. The proportion of Firmicutes(88.60%)was the most. Only 6 genera were observed at high abundance (detection rate >1%). These included: Streptococcus (70.28%), GranuIicatella (6.20%), Gemella (3.10%), Veillonella (1.81%), Neisseria (1.03%) and Haemophilus (1.03%). Among them, Streptococcus was detected in all 8 samples. Conclusion: The salivary microbial community structure of healthy children in Bonan is more abundant. Streptococcus is the dominant bacteria in saliva of healthy children in Bonan, and its specific mechanism of maintaining oral health status in children may need a further study.
