重组人p53腺病毒的应用前瞻

恶性肿瘤的发生与基因突变有密切关系。抑癌基因p53编码转录因子P53蛋白在调控癌基因表达和抑制血管增生、参与细胞周期调控及凋亡、DNA合成和RNA的转录及多药耐药清除中起重要作用,与肿瘤的发生、发展及预后关系密切。已证实50%以上肿瘤细胞存在p53基因突变、易位、融合、插入或缺失。p53基因治疗的目的是导入野生型p53,诱导肿瘤细胞凋亡、纠正瘤细胞中突变的p53基因功能、增强化放疗敏感性,在多种实体瘤的体内外试验中显示出良好疗效。本综述就这一领域所取得成绩及目前所存在的问题做了进一步阐述,以便更好、更广泛的应用于临床。

Malignant transformation is closely related to gene mutations. The tumor suppressor gene p53 codes for a transcription factor, P53 p rotein, plays a critical regulation role in oncogene expression, inhibinting vascular proliferation,DNA synthesis and repair systems, cell apoptosis and multidrug resistance. It has been demonstrated that p53 mutation,translocation,fusion,insertion and deletion occurs in more than 50% of cancer cells. which is thought to play a crucial role in malignant transformation. p53 gene therapy approaches aiming at a direct correction of the specific molecular defect in p53-mutated malignant cells and enhancing sensitivity of chemtherapy and radiotherapy. In this review, we discussed the current advances and problems in this field, focusing on adenovirus-mediated wild-type p53gene therapy.
