GDF11(growth differentiation factor 11,GDF11)是TGF-β超家族的成员之一,既往研究表明GDF11可在衰老过程中调节大脑、骨骼肌、心肌的再生,而GDF11在骨代谢中的作用机制不明。在本研究中,通过分离培养骨髓基质干细胞,研究 GDF11作用信号通路,发现GDF11通过诱导SMAD2/3磷酸化抑制Runx2表达,从而抑制骨髓机制干细胞向成骨细胞分化。研究结果解析了GDF11调节骨代谢机制,为治疗老年性骨质疏松症提供潜在治疗靶点。

Growth Differentiation Factor 11 (GDF11) is one of the members of the TGF-β superfamily. Recent studies confirmed that GDF11 plays important role to regulate the regeneration of brain, skeletal muscle and heart in aging. However, the action of GDF11 on bone metabolism remains unclear. In the present study, our results showed that GDF11 inhibited the osteoblast differentiation of bone marrow mesenchymal stem cells (BMSCs) in vitro. Mechanistically, GDF11 repressed Runx2 expression via inducing SMAD2/3 phosphorylation during the process of osteoblast differentiation. These results provide mechanistic insight into how GDF11 regulates bone mass, while also providing a further rationale for the therapeutic targeting to treat age-related osteoporosis.
