产β-甘露聚糖酶菌株的筛选及产酶条件的优化
Selection Strains for the production of β-Mannanase and Optimization of Fermentation Conditions
目的:从堆肥、热泉等高温环境中筛选产β-甘露聚糖酶真菌菌株,优化产酶条件,为工业化发酵生产奠定理论基础。方法:设计培养基,通过富集和选择培养,分离筛选产β-甘露聚糖酶的真菌菌株;利用形态学和分子生物学对菌株进行鉴定;通过液体发酵,测定产酶活性,优化产酶条件。结果:获得9株具有降解β-甘露聚糖能力的真菌菌株,其中菌株PSC-D-G18降解能力最强,产酶活性为56 U/ml,其最适产酶pH为5,最适温度条件为60℃。菌株鉴定结果表明,该菌株为申克氏孢子丝菌(Sporothrix.schenckii)。结论:菌株PSC-D-G18产β-甘露聚糖酶活性较强,具有潜在的应用价值。
Objective: To select strains for the production of β-Mannanase from compost, hot springs and other high temperature environment and optimize conditions of enzyme production for the industrialization. Methods: Designing enrichment and selection medium in order to select the fungal strains with β-mannanase; Identification those strains by morphological and molecular biology methods; Determining production activity and optimizing conditions of the enzyme production by liquid fermentation. Results: 9 fungal strains with β-mannanase were obtained, including the strain of PSC-D-G18 which had the stronger enzyme activity that was 56 U/ml. The optimum pH and temperature for the β-mannanase activity were 5.0 and 60℃, respectively. This fungal strain was identificated as Sporothrix.schenckii. Conclusion:The strain of PSC-D-G18 which had the stronger enzyme activity of β-mannanase that could be exploited as a potential source of feed industry.
