目的:探讨凋亡抑制基因survivin 在视网膜/脉络膜血管内皮细胞(RF/6A)中的作用。 方法:应用免疫荧光的方法检测survivin基因在RF/6A细胞内的表达以及定位。通过siRNA干扰技术降低survivin基因在RF/6A细胞中的表达,用RT-PCR检测mRNA表达变化,用Western Blot检测蛋白表达变化。采用MTT,Boyden小室,免疫荧光,流式细胞仪对细胞的增殖,迁移,伸展,细胞周期和凋亡变化进行检测。用Matrigel对RF/6A血管形成进行检测。 结果:RF/6A细胞中survivin 阳性表达于细胞核。siRNA干扰技术在mRNA水平以及蛋白水平均有效的沉默survivin基因在细胞中的表达。survivin siRNA干扰后RF/6A细胞的增殖、迁移均受到抑制,细胞在缺氧的条件下凋亡明显增加,RF/6A血管形成明显受到抑制,而细胞的粘附和伸展能力未受明显影响。 结论: Survivin siRNA对RF/6A细胞的细胞增殖、迁移能力的抑制作用以及凋亡的促进作用明显。Survivin 基因为临床玻璃体视网膜血管性增殖性疾病的治疗提供了新的靶点。

Purpose: To evaluate the roles of survivin in the retinal/choroidal vascular endothelial cells (RF/6A). Methods: Immunofluorescence analyses were performed to determine the sites of survivin in RF/6A cells. siRNA technology was used to knock down survivin expression and to study its effects on cells in vitro. Cell proliferation, migration, spreading, cycling, and apoptosis were assessed with the MTT assay, Boyden chamber assay, immunocytochemistry, and flow cytometry. Tube formation by RF/6A on Matrigel was also analyzed. Results: Survivin was expressed in the nucleus in RF/6A cells in vitro. Survivin siRNA knockdown inhibited cell proliferation, migration. Under hypoxic conditions, more apoptotic cells were observed among the knockdown cells than among the control cells.Tube formation by RF/6A cells was also disturbed. Conclusions: Silencing the expression of survivin in RF/6A cells inhibited their proliferation and reduced their tolerance of hypoxic conditions. This study indicates that survivin may be a molecular target for new diagnostic and therapeutic strategies for Proliferative vitreoretinopathy diseases.
