为了解栽培丹参rDNA内转录间隔区(ITS)的遗传多样性、其系统发育关系和寻找其单核苷酸多态性(SNP)指纹用以准确快速鉴别丹参栽培居群,本研究采用PCR扩增了国内30多个地区的42个丹参居群的ITS序列并进行了比较分析。结果表明:丹参rDNA的ITS1、5.8SrDNA和ITS2区的长度分别为203~229bp、164bp和226~229bp;其ITS1区大小变化较大,而ITS2区核苷酸变异大。其SNP变异位点存在于ITS1和ITS2区,变异位点数分别为 19和37个,变异率分别为8.30%和16.16%。基于ITS 的SNP指纹只能鉴别3个栽培丹参居群;ITS系统发育树显示, 丹参栽培居群之间的亲缘关系与地理来源关系不大。

o understand the genetic diversity of the cultivated Salvia miltiorrhiza populations, the phylogenetic relationship based on the internal transcribed spacer (ITS) regions of the rDNA, and to find the single nucleotide polymorphism (SNP) fingerprints for the identification of S. miltiorrhiza populations, this study PCR amplified, sequenced and compared the ITS sequences of the 42 S. miltiorrhiza populations collected from 30 different regions in China. Results showed that the lengths of ITS1, 5.8S rDNA and ITS2 regions in rDNA of S. miltiorrhiza were 203-229bp, 164bp, and 226-229bp respectively; relatively larger difference in length existed in ITS1, while more nucleotide variations occurred in ITS2; SNP variation sites occurred only in the ITS1 and ITS2 regions with 19 and 37 variation sites and 8.30% and 16.16% variation rates respectively. Based on the SNP fingerprints of ITS, only 3 cultivated populations could be discriminated from the rest. The phylogenetic trees based on various regions of ITS showed that the cultivated S. miltiorrhiza populations were not genetically related to their geographical sources.
