电针对慢性应激抑郁大鼠海马激活素A和晚期糖基化终末产物受体表达的影响

目的 观察电针对慢性应激模型大鼠海马激活素A (activin A,ACT A) 和晚期糖基化终末产物受体(receptor for advanced glycation end products ,RAGE)蛋白表达的影响,探讨电针对慢性应激抑郁模型大鼠海马神经元的抗损伤和促生长作用。方法 将40只雄性SD大鼠随机分为四组:空白组、模型组、模型+电针组(以下简称电针组)、模型+氟西汀组(以下简称氟西汀组)。空白组正常饲养28天,模型组、电针组、氟西汀组均采用慢性应激结合孤养方法造模28天。电针组于造模前1 h,选取"百会"、"印堂"穴进行电针治疗,频率2 Hz,电流强度0.6 mA,每次 20 min;氟西汀组于造模前1 h,给予盐酸氟西汀混悬液5 mlokg-1灌胃治疗。通过生物素标记抗体芯片技术筛选出与神经元生长和损伤相关的差异蛋白ACT A、RAGE。结果 与空白组比较,模型组大鼠海马ACT A和RAGE蛋白表达均上调(fold change=1.240,1.356);与模型组比较,电针组、氟西汀组ACT A蛋白表达均下调(fold change=0.701,0.604)、RAGE蛋白表达均下调(fold change=0.617,0.624)。结论 电针通过下调RAGE蛋白表达水平以发挥神经元抗损伤作用,并且良性调节具有神经保护作用的ACT A,使其趋于正常水平,揭示电针可能具有促进神经元再生的作用,这些可能是针刺抗抑郁作用的相关分子机制之一。
Objective To explore the adjustment of electro-acupuncture on the expression of proteins of activin A (ACT A) and receptor for advanced glycation end products (RAGE) in the hippocampus of rats with chronic unpredictable mild stress-induced depression;To investigate the Anti-damage and growth promoting effect of electro-acupuncture on the the protection of hippocampal neurons. Methods 40 Sprague-Dawley rats were randomly divided into four groups: control group,model group,model+Electro-acupuncture group (EA group),and model+fluoxetine group (fluoxetine group). control group were raised for 28 days without any stimulation and treatment, the other groups were modeling by the chronic unpredictable mild stress (CUMS) combined with solitary raising methods to establish rat depression model for 28 days.EA group was given acupuncture on acupoints Baihui (DU 20) and Yintang (DU 29) for twenty minutes(2 Hz,0.6 mA)before modeling.Fluoxetine group was given fluoxetine at the dosage of 5 mlokg-1 before modeling. The expression of proteins of Act A and RAGE in hippocampus of rats were detected by biotin label-based antibody array.Results Compared to the control group,the expression of protein of Act A and RAGE ware both upregulation (fold change=1.240,1.356) in the model group. Compared to the model group, the expression of protein of Act A and RAGE ware downregulation (fold change=0.701,0.604; fold change=0.617,0.624).conclusion The treatment of electro-acupuncture could down- regulate the protein expression level of RAGE in order to resist damage damage and benign-regulate the protein expression level of Activin A to normal level in the hippocampus of rats with chronic unpredictable mild stress-induced depression, which may be involved in promoting the regeneration of neurons. All of it may be one of the molecular mechanism of antidepressant effect of electro-acupuncture.
