目的:旨在研究BCL2L11表达异常在胰腺癌发病中的调控机制,并探索miRNA-BCL2L11信号通路对胰腺癌增殖、凋亡等生物学行为的影响。方法:通过生物信息学软件筛选出BCL2L11可能是miR-24的靶基因,利用荧光素酶报告基因验证miR-24与BCL2L11的调控关系,裸鼠腋下植瘤(Panc-1细胞过表达BCL2L11组、过表达miR-24组、对照组),4周后处死小鼠,取出肿瘤。测量肿瘤大小及重量,检测组织中BCL2L11及 miR-24表达水平。及对血管生成的调控。结果:荧光素酶报告基因结果显示miR-24可直接与BCL2L11的3'UTR区结合,miR-24过表达组的肿瘤生长较快,并且其BCL2L11蛋白表达水平明显下降,但mRNA水平无明显变化。MiR-24可以通过抑制BCL2L11的表达抑制体内肿瘤生长。结论:证实miR-24通过抑制BCL2L11表达调控胰腺癌细胞的增殖和凋亡。

his study aimed to explore the expression of BCL2L11 in Pancreatic cancer,select miRNAs which directly target BCL2L11 and further discuss the role of the miRNA-BCL2L11 pathway in the tumorigenesis ofin PaC. By using bioinformatics tools, we found that miR-24 can directly target the 3'UTR of BCL2L11 mRNA. Panc1 cells treated with control lentivirus or miR-24 overexpressing lentivirus or BCL2L11 overexpressing lentivirus were injected subcutaneously into nude mice. Mice were sacrificed after 4 weeks, and the weight and diameter of tumors were recorded. The expression of BCL2L11 and miR-24 were determined thereafter. ?????
