分离测定鱼腥草和山楂果实中槲皮素、芦丁和绿原酸反向迁移胶束毛细管电泳新方法
Separation and determination of quercetin, rutin and chlorogenic acid in herba houttuyniae and hawthorn fruit by reverse-flow micellar electrokinetic chromatographic
本文建立了反向迁移胶束毛细管电泳法(RF-MEKC)同时分离测定鱼腥草和山楂果实中槲皮素、芦丁和绿原酸的新方法。运行缓冲溶液为70 mM 十二烷基硫酸钠(SDS)-30 mM 磷酸二氢钠(NaH2PO4)、pH 2.1,分离电压-15KV,检测波长214 nm。在此最佳条件下,分 析物可在15 min内实现基线分离。槲皮素、芦丁和绿原酸的线性范围分别为25-400 μg/mL、25-400 μg/mL和12.5-200 μg/mL (r=0.9992、r=0.9992、r=0.9999),三种分析物的检测限(S/N=3)分别为2.52、3.10、3.64 μg/mL。槲皮素的回收率为98.5%-103.7%,芦丁的回收率为95.8%-97.6%、绿原酸回收率为99.1%-103.8%。
A reverse-flow micellar electrokinetic chromatographic (RF-MEKC) method with direct on-column UV detection was developed for the separation and determination of quercetin, rutin and chlorogenic acid in herba houttuyniae and hawthorn fruit. The separation conditions of the three analytes were optimized with respect to the concentrations of SDS, NaH2PO4, pH and applied voltage. Baseline separation was obtained for the three analytes within 15 min, using a running buffer containing 70 mM SDS, 30 mM NaH2PO4, pH 2.1 and −15 kV voltage. The linear calibration range was 25-400 µg/mL for quercetin and rutin; 12.5-200 µg/mL for chlorogenic acid (r=0.9992、r=0.9992、r=0.9999). The recoveries were 98.5%~103.7%, 95.8%~97.6% and 99.1~103.8% for quercetin, rutin and chlorogenic acid, respectively. The detection limits of three analytes (S/N=3) were 2.52、3.1、3.64 μg/mL, respectively.
