目的] 研究血清剥夺条件下雌激素对原代心肌细胞活力的影响。[方法] 通过MTT法测定细胞活力,western blot技术测定蛋白表达。[结果] 血清剥夺培养条件下心肌细胞活力下降,细胞死亡增加。血清剥夺条件下加入雌激素,雌激素加重血清剥夺诱导的心肌细胞死亡,此作用呈浓度依赖性。雌激素对正常培养的心肌细胞的细胞活力无影响。血清剥夺不同培养时间,均诱导心肌细胞LC3II表达增加,即血清剥夺诱导心肌细胞自噬反应增加。雌激素可抑制血清剥夺诱导的心肌细胞自噬反应增加,而雌激素对心肌细胞的基础自噬水平无影响。通过雷帕霉素诱导心肌细胞自噬,进一步观察了细胞自噬是否参与雌激素的损伤作用,结果显示雷帕霉素增加细胞自噬,但自噬的诱导并未逆转雌激素在血清剥夺条件下加重心肌细胞死亡的作用。在不同的血清剥夺培养时间内,血清剥夺均引起p-smad1/5/8水平下降;骨成型蛋白-2(BMP2)处理可逆转血清剥夺诱导的p-smad1/5/8水平下降,并可抑制血清剥夺诱导的心肌细胞活力下降。[结论] 雌激素加重血清剥夺诱导的原代心肌细胞死亡;骨成型蛋白-2减轻雌激素对血清剥夺诱导的心肌细胞损伤加重作用。

Object] To study the effects of estrogen on the cell viability of cardiomyocytes cultured with serum deprivation. [Methods] The cell viability of cardiomyocytes was determined by MTT assay and the protein expression was determined by western blot. [Results] Cell culture with serum deprivation decreased cell viability of cardiomyocytes and increased cell death. Estrogen treatment aggravated serum deprivation-induced cardiomyocyte death in a dose-dependent manner. Estrogen showed no effect on cardiomyocyte viability in normal culture conditions. Serum deprivation for different time induced increase of LC3II expression, indicating that serum deprivation induced increase of cardiomyocyte autophagy. Estrogen treatment inhibited the autophagy response induced by serum deprivation. Rapamycin treatment induced increase of cardiomyocyte autophagy, but rapamycin did not alleviate the effect of estrogen-aggravated serum deprivation-induced cardiomyocyte death. The level of p-smad1/5/8 was decreased in cultured cardiomyocytes with different serum deprivation. Bone morphogenetic protein-2 (BMP2) alleviated serum deprivation-induced decrease of p-smad1/5/8 expression and alleviated serum deprivation-induced decrease of cell viability. [Conclusions] Estrogen aggravated serum deprivation-induced cardiomyocyte death; Bone morphogenetic protein-2 alleviated estrogen-aggravated serum deprivation-induced cardiomyocyte death.
