红枝垂组织培养快繁技术的研究

Study on Tissue Culture and Rapid Propagation technology of C. subhirtella var. pendula ‘Rosea’

本文以红枝垂(C. subhirtella var. pendula ‘Rosea’)的带芽茎段为外植体,接种到MS+1.0 mg/L BA+0.1 mg/L NAA培养基上,芽的诱导率最高,为80%;采用正交设计方法,研究了6-BA、NAA、蔗糖3种不同因素及其组合对红枝垂芽增殖系数的影响,结果表明:细胞分裂素BA是幼芽增殖的最主要因素。芽增殖的最佳组合为A2B3C2即MS+0.5 mg/L BA+0.1 mg/L NAA+35 g/L 蔗糖,其增殖系数为4.15。当株高在2-3 cm时,转入1/2MS+1.0mg/L NAA +1.0mg/L IBA生根培养基上,接种25d后,生根率可达95%;经过5-7d的开瓶炼苗,以沙:珍珠岩:蛭石=1:1:1为基质,20d后调查,其移栽成活率达84%。

Selected explants, stem section with bud,were inoculated medium of MS 1.0 mg /L N6 -benzyladenine (BA) and 0.1mg/L α-naphthaleneacetic acid (NAA) , the highest shoot induction rate is 80%. The N6 -benzyladenine (BA), α-naphthaleneacetic acid (NAA) and sucrose were used to compare their effect on shoot proliferation coefficient in the C. subhirtella var. pendula ‘Rosea’ by using the orthogonal experiment design. The test result indicated that cytokinins(BA) was the main factor. The optical medium for reproduction of the bud was A2B3C2, namely, MS with 0.5 mg /L BA and 0.1 mg /L NAA + 35 g/L sucrose; Proliferation coefficient was 4.15. Plantets were transplanted the 1/2MS medium with 1.0 mg/L indole-3-btyricacid (IBA) and 1.0mg/L α-naphthaleneacetic acid (NAA) for rooting, when the explants were reached 2-3cm.,the rooting rate could reach 95% after 25 days. Opening the culture bottle during seedling training in 5-7d, Substrate with sand, perlite and vermiculite by 1 to 1 to 1 was the optimal. After 20 days the survival rate of transplants reached 84%.
