小鼠皮肤切创愈合过程中CB1受体的变化规律及其与损伤时间相关性的研究
he Time-dependent Changes of CB1R Expression during the Skin Incised Wound Healing in Mice
目的 探讨小鼠皮肤切创愈合过程中损伤区CB1受体的变化及不同时间的CB1受体的变化规律。方法 应用免疫组织化学和Western blot 方法检测小鼠皮肤切创后不同时间段CB1受体的变化情况。 结果 正常组织中CB1受体表达于表皮、毛囊、皮脂腺及皮肌层细胞。伤后6h~12h,未见多核粒细胞呈CB1受体阳性染色。伤后1d~5d,阳性染色以单核细胞和成纤维细胞为主。伤后7~14d,CB1受体阳性着色以成纤维细胞为主。阳性细胞率由伤后6h至3d逐渐升高,5d达到高峰,7d~14d逐渐降低。Western blot 检测显示,各个时间段均有CB1受体的阳性条带,其中伤后5d为CB1表达的高峰期。结论 在小鼠皮肤损伤愈合过程中CB1受体在单核细胞及成纤维细胞中表达,可能参与皮肤损伤后的愈合。其在伤后不同时间的表达变化规律可用于皮肤损伤时间的推断。
Objective To investigate the change of cannabinoid receptor type 1 (CB1R) during the the skin incised wound healing in mice and to explore the rule of the time-dependent change of CB1R in wound age determination. Methods The changes of CB1 expression in incised skin wound were detected by immunohistochemistry and Western blot. Results There was a minimal baseline staining of CB1 in control mouse skin,which is expressed in epidermis, hair follicles, sebaceous gland, muscular layer. Changes of CB1 expression were undetectable in neutrophils in the wound specimens from 6 hours to 12 hours after injury. The CB1 positive cells were mostly mononuclear cells and fibroblasts between post-injury day 1 and 5, whereas the CB1 postive cells were mostly fibroblasts between post-injury day 7 and day 14. Morphometrically, the ratio of the CB1 positive cells to the total increased gradually in the wound specimens from 6h to day 3, maximized at day 5, and then decreased guadually from post-injury day 7 to day 14. The changes of CB1 expression were observed throughout the wound healing stages by Western blot as well, with a peak in specimens of day 5. Conclusion CB1 was activated during the wound healing.It is partly expressed in mononeuclear cells, which may be involved in inflammation. CB1 was observed in fibroblasts, which may participate in wound healing, and meanwhile, CB1 may be a potentially useful marker for wound age determination.
